New Magnetic Nano Fe3O4 Gene Vector Improve Adenovirus vector's Efficicency

Objectives: We try to develop a new magnetic nano gene vector to improve the efficicency and target of transgene therapy for salivary tumor.

Methods: We used surface-initiated Atom Transfer Radical Polymerization(ATRP) reaction to prepare Fe3O4@PDMA+ Core-shell Nanoparticles used as magnetic nano gene vector which surface contain positive charge.Then Fe3O4@PDMA+ core-shell nanoparticles mixed with AdCMV-EGFP and they combined by electrostatic interation .We transfected the complex to SACC-83 cell with magnetic field to improve the transfection efficicency and used the AdTERT-EGFP as control.We used Fluorescence inverted microscope to observe the EGFP expression and flow cytometry to detect the transfection efficicency.

Results:The monodisperse Fe3O4@PDMA+ core-shell nanoparticles were synthesized and the particles have a narrow size distribution about 6nm. TEM micrographs show the particles contained a thin layer of polymer shell.The Fe3O4@PDMA+ core-shell nanoparticles were well dispersed in water. The data of flow cytometry show the transfection efficicency improve compare with the control group(P<0.05).

Conclusion:We succeed synthesis a new magnetic nano gene vector and it improve the Ad gene vector's efficicency.This will lower the dosage of Ad vector applicated and the toxicity too.Our research achievements provide a foundation for us to investigate the target transgene ability of magnetic nanoparticle in vivo and in salivary tumors, as well as theory basis and practice experience for us to conduct gene therapy clinically with high efficiency, targeting effect and no trauma, which will develop a new way to treat salivary tumors.
