Gene Expression Profiles of Dendritic Cells from Mucosa and Skin

Objectives: Dendritic cells are one of the most potent antigen-present cells (APC) for activation of mucosal and systemic immune responses. This study is to analyze the difference of gene expression of dendritic cells from Peyer's patches (mucosal) and langerhans cells from skin (systemic). Methods: We used magnetic beads and flow cytometric sorter to isolate dendritic cells from Peyer's patches and Langerhans cells from skin. Then, mRNA of the samples was isolated and applied to microarray hybridization. Candidate genes were picked out from microarray data analyses and examined by real-time reverse transcriptase polymerase chain reaction (real-time PCR). Results: We revealed that 3548 genes fold change were ≥2. 114 genes were related to immunity system in according to gene ontology. 28 genes of these 114 genes were selected and confirmed their gene expression by real-time PCR. Conclusions: The list of 28 genes was valuable candidates to be further research the difference of dendritic cells from mucosa and skin.
