In vitro study of salivary gland cells in sialolithiasis

Objectives: The aim of this research was to study some issues of etiology, chemical composition and structureof sialolithiasis. Study materials and method: Totally 50 micro samples were prepared and investigated with confocal laser scanning microscope (10 X 40,Zeiss LSM Pascal). The cells of human submandibular gland (HSG) and parotid gland (HSY) were cultured, infected/ treated/ with JFP and BMP-2 adenovirus, and then we defined the alkaline phosphatasa, type 1 collagen, osteocalcin, osteopontin gene expession level in the infected parotid gland cells and compared them with those in non- infected cells.

Study results: By the investigation were found massive calcifications, fat tissue infiltrate, decrease of acinis cells, fibrous atrophy of adenoid epithelium in the salivary gland tissue under microscopy.

By the Polyacrylamyde Gel Electrophoresis (PAGE) conducted on proteins extracted from BMP-2 adenovirus infected HSY salivary gland cells the type 1 collagen , osteocalcin, osteopontin were determinated. By the staining method in non- infected cells the level of alkaline phosphatasa activity was zero or low; on other hand, in the infected /treated/ cells it was positive or had high activity level.

Conclusion:

1. Massive calcification, decrease of acinis cells number, fat infiltration and replacement of parynchematous tissue by the fibrous are common in sialolithiasis formation in submandibular salivary gland

2. Gene expressions of type 1 collagen, osteocalcin, osteopontin, alkaline phosphatasa were relatively high in adenovirus infected / treated/ HSY salivary gland cells.
