New method for evaluating elimination-rate of protein-I/II(streptococcus-mutans) by different irrigants

Objectives:The aim of this study was to determine the effectiveness of eight endodontic irrigant solutions used with a conventional system to eliminate oral streptococcus mutans protein I/IIf from inoculated root canals. Destilled water, lauryl sulfonate, mercryl lauryl, 2.5 % sodium hypochlorite, 2 % chlorhexidine and 3 % hydrogen peroxide have been used as antimicrobial agents.

Methods: PCR, cloning, radioactivity analysis (computerized gamma counter).

Results: Sodium hypochlorite solution with ultrasonic flushing was statistically more significant in removing Streptococcus mutans protein I/IIf inoculated in root canals. The solutions without sodium hypochlorite demonstrated less effectiveness in eliminating protein I/IIf.Quantitative analysis obtained from this study demonstrated the efficacy of the radioisotop method used to evaluate the effectiveness of different endodontic irrigant solutions. Furthermore an analysis of the topographic spread out architecture of the pathogen is also possible. Conclusion: The technique used has a high specificity and sensitivity to quantify the pathogens of interest and could provide new insights into the possible pathogenic mechanisms of the pathogens of interest throughout the identification of their localisation. It can also be used for measuring the effectiveness of solutions and agents with antimicrobial activity.
