Pyrophosphate and SIBLING Proteins Regulating Mineralization

Description: Molecular determinants of skeletal, dental and pathologic mineralization include inhibitory SIBLING proteins, and the small-molecule inhibitor pyrophosphate. These mineralization inhibitors are substrates for tissue-nonspecific alkaline phosphatase, an enzyme that degrades pyrophosphate and dephosphorylates SIBLING proteins, thus modifying their mineral-inhibiting activity. The influence of these factors on mineralization will be discussed.
